چکیده

L-Type amino acid transporters such as LAT1 and LAT3 mediate the uptake of essential amino acids. Here, we report that prostate cancer cells coordinate the expression of LAT1 and LAT3 to maintain sufficient levels of leucine needed for mTORC1 signaling and cell growth. Inhibiting LAT function was sufficient to decrease cell growth andmTORC1 signaling in prostate cancer cells. These cellsmaintained levels of amino acid influx through androgen receptor–mediated regulation of LAT3 expression and ATF4 regulation of LAT1 expression after amino acid deprivation. These responses remained intact in primary prostate cancer, as indicated by high levels of LAT3 in primary disease, and by increased levels of LAT1 after hormone ablation and in metastatic lesions. Taken together, our results show how prostate cancer cells respond to demands for increased essential amino acids by coordinately activating amino acid transporter pathways vital for tumor outgrowth. Androgen receptor and nutrient signaling pathways coordinate increased amino acid transport in prostate cancer progression

Background Solid tumors including prostate cancer activate angiogenic signals to ensure an adequate blood supply. In parallel, amino acid transporters on the cell surface are also increased so as to provide nutrients for the higher metabolic and growth demands of cancers. We are studying the L-type amino acid transporters (LAT1 and LAT3) that mediate uptake of essential amino acids including le...

Molecular and Cellular Pathobiology Collaboration of Kras and Androgen Receptor Signaling Stimulates EZH2 Expression and Tumor-PropagatingCells in Prostate Cancer

Elevation of the chromatin repression factor enhancer of zeste homolog (EZH2) is associated with progression and poor prognosis in several human cancers including prostate cancer. However, the mechanisms driving EZH2 expression are not fully understood. In this study, we investigated the functional synergy in prostate cancers in mice resulting from activation of the androgen receptor, Kras, and...

Targeting inflammatory cytokines-androgen receptor (AR) signaling with ASC-J9® to better battle prostate cancer progression

Inflammatory cytokines and chemokines released by macrophages in the prostate cancer microenvironment may signal via the androgen receptor (AR) to influence tumor progression. In particular, macrophages appear to promote tumorigenesis by altering the chemokine (C-C motif) ligand 4 (CCL4)/AR signaling axis. This process can be blocked by ASC-J9®, an enhancer of AR degradation. ASC-J9® also inhib...

The Role of Cyclooxygenase-2 in Signaling Pathways Promoting Colorectal Cancer

Colorectal cancer is one of the most common cancers in the world. Various factors are involved in the development and progression of this disease. One of these agents is cyclooxygenase-2 (COX-2). COX-2 is a product of the PTGS2 gene and converts free arachidonic acid to prostaglandins. COX-2 is not naturally expressed in most normal cells. Noticeably, the increased expression of COX-2 has been ...
